Pyro QC Pak

Microplates (also called “microtitre,” “microtiter ®,” or “micro titration” plates) provide a rapid way to analyze large numbers of tests. Samples are typically on the order of a few hundred µL, and a large number of plates can be measured in a short period of time. A 96 well plate is commonly used for these analyses, however 12, 24, 48, 384, and 1536 well plates are also available.

A broad variety of analytical measurements can be performed in microplates including absorbance (OD), luminescence (L), fluorescence intensity (F), time-resolved fluorescence (TRF), and fluorescence polarization (FP). In each case, the fundamental measurement is the same as when a standard cuvette is used, but the instrument (called a “reader”) is capable of reading the entire plate is less than a minute and generate a complete report.

While the general design of a reader is identical to that of a cuvette system (e.g. an absorbance reader is similar in nature to a spectrometer), the optics are configured such that 96 samples can be read on a plate with an 8 x 12 format.

Basics

  • Evaporative cooling : The (cooling) effect of evaporation may be included in the measurement by adding distilled water to the well positions. Note: the local humidity will influence evaporation and therefore the temperature. Movement of the plate will influence the local humidity.
  • Thermal delay of the TTP : The design of the TTP approximates the mass of a real microplate, so that warm-up and cooling times are similar those observed using a standard microplate. Typical time for heating from RT to 37°C is about 45 minutes using non-contact heating.
  • Each sensor (one measurement position) measures the average of 4 wells.

Modes

Measurement without liquid:

For routine testing of incubators and temperature controlled instruments, especially incubating photometers, fluorometers, and luminometers.

Measurement with liquid (distillated water)

Plate uncovered

This measurement mode is intended for instruments in which the microplates are normally used uncovered.

Covered plate

This measurement mode is intended for instruments in which the microplates are normally covered, such as incubators used in tissue culture applications.


User Controls:

Power Switch:

  • The power must be switched on, for temperature measurements and PC communication.
  • For charging, the TTP can be on or off .
  • Powering off the TTP will stop a measurement, in which case the data will be retained.

Green Start button:

Begins a new measurement procedure.

Green LED:

  • When lit, the plate is ready for measuring.
  • When flashing, the measurement is finished and the memory is full. (Download the data to PC and clear the memory, in order to continue.)
  • Pushing the green start button begins a new measurement, and the old measurement will be cleared.
  • Green led switched off: measurement is running (or TTP is switched off)

Red LED: when lit, the battery is low ( <3.0V )

!Caution: below 2.8V, a security switch automatically powers off the TTP!
